4.7 Gas supply
4.7.1 Gas mixture required
WARNING: Select the appropriate high-purity gas mixture to suit the
altitude and the culture media that is used.
IMPORTANT NOTE: To maintain the correct operating pH level in
bicarbonate-buffered media, the concentration of atmospheric CO
contact with the media must be strictly controlled.
When the Cook Culture System is used at sea level, a 6% CO
89% N
blend high-purity gas mixture is recommended.

Cook Medical recommends reducing the oxygen concentration from the
normal atmospheric concentration to a value of 5%-8% in the atmosphere
that is in contact with the media, because this reduced concentration may
reduce the formation of reactive oxygen species.
It is recommended that the end user perform in-situ testing to
determine the correct gas mix that is needed to attain the desired
pH of the IVF media.

4.7.2 Gas regulator selection
The following information contains recommendations for the selection
of the gas cylinder regulator that is to be used with the MINC+. Following
these guidelines will provide a reliable gas connection between a gas
cylinder and the MINC+.
IMPORTANT NOTE: The MINC+ incubator must be connected to a
regulated gas supply with a delivery pressure of 150 kPa ± 15 kPa at the MINC+
gas inlet. It cannot be directly connected to an unregulated gas supply. The
gas regulator is an important part of the gas delivery system, and it must be
designed and manufactured to supply medical-grade gases that are highly
pure and accurately delivered to the device. It is also important that the
regulator does not contaminate the gas stream.
Please request these features when ordering your gas regulator:
• Single-stage high-purity regulator.
• Metal-to-metal diaphragm seal.
• Stainless steel diaphragm, because stainless steel does not contaminate
high-purity gas streams.
