4 The closed green circle indicates that the calibration is completed. After all calibrations are completed, the
instrument can be switched into acquisition mode.

Volume calibration
After exchange of the sample tubing, the system must be calibrated to this specific tubing in order to correctly
set the tube volume and flow rate. This process is automated as an Express Mode VolumeCalibration using
MACSQuant Calibration Beads. To ensure accurate cell count, repeat the volume calibration once a month. The
process takes ~48 minutes.
It is recommended to run buffer as sample to prepare the system, e.g. 3× 100 µL MACSQuant Running
buffer with high flow rate.
1 Go to the Experiment tab.
2 Go to the Settings tab.
3 Select the Express Mode radio button.
4 Select Setup from the Type drop-down menu.
5 Select VolumeCalibration from the Mode drop-down menu.
6 Select Single tube rack from the Rack drop-down menu.
7 Place an empty 5 mL tube in the Single tube rack.
8 Ensure that the buffer bottle is full and the waste bottle is empty.
9 Click the Start button.
10 Follow the on-screen instructions.
