If the rack type is not displayed use the next or previous window button in the icon bar to switch to
another window.
3 Click Start calibration under the heading Custom 96 rack or Custom 384 Well Rack. The robotic needle
arm will automatically move forward.
4 Proceed as described for standard 96 or 384 well plates. Refer to Calibrating the Chill 96 Rack on
page 61 or Calibrating the 384-well plate on page 61, respectively.

Calibrating the Reagent Rack
1 The MACSQuant Instrument can be used in combination with both the Universal Reagent Rack and the
Reagent Rack 4. The instructions for calibration apply to both.
2 Go to the Tools tab in the side panel of the MACSQuantify Software and select the Calibrate uptake unit
box. Red closed circles indicate that the uptake components are not calibrated.
3 Place the Universal Reagent Rack onto the MACSQuant X Orbital Shaker.
4 Place an empty reagent vial on position 4 (front-most position) of the Universal Reagent Rack.
5 Remove the cap from the reagent vial.
6 Click Start calibration under the heading Reagent Rack.
7 Select a reagent rack from the drop-down menu.
8 A dialog box will prompt you to ensure that the Universal Reagent Rack is installed correctly and that the
Single tube rack is removed.
9 Confirm the dialog box by clicking OK. The robotic needle arm will automatically move forward.
10 Adjust the uptake needle to the deepest point of the vial on position 4.
11 Lower the robotic needle arm carefully until it touches the bottom of the vial.
12 Click Save calibration. The closed green circle indicates that the calibration is completed.
13 Click Test to confirm successful calibration. The MACSQuant Instrument tests all positions.

Calibrating the Single tube rack detection
Activation of the Single tube rack detection avoids a collision of the MACSQuant X Orbital shaker with a
mounted Single tube rack.
1 Click on the Tools tab in the side panel of the MACSQuantify Software and select the Calibrate rack
detection box. Red closed circles indicate that the components are not calibrated.
2 Click Start calibration under the heading Single tube rack detection.
