  • Page 12 External laser devices connected to to the RS232/BCR port have to comply with the standard IEC 60825-1. In order to ensure electromagnetic compliance, connect peripheral devices only with double-shielded cables and only use the power cable and the keypad included in delivery. Use the keypad with MACSQuant X models only.
  • Page 13 Optical radiation hazards The MACSQuant X is classified as a Class 1 M laser product per standard IEC 60825-1. The MACSQuant X is equipped with three Continous-Wave Lasers (laser class 3B). These lasers are secured by a protective housing. Do not remove the protective housing. Use of controls or adjustments or performance of procedures other than those specified herein may result in hazardous radiation exposure.

  • Page 53 CAUTION! Two persons are required to lift the instrument. Take care to avoid personal strain or injury. The instrument is heavier at the front and should be stabilized while lifting it.   • Page 54 Assembly 2.6.1 Install the fluid bottles Operating the instrument requires Running Buffer, Washing Solution, and Storage Solution. One bottle is required for waste collection. Always operate the instrument with ready-to-use MACS Buffers and solutions only. The bottles, closures, tubing and fluid sensor cables are color-coded for easier handling. 1 Install only one fluid bottle at a time.

The Running Buffer and the Waste bottle lids have each two connectors for tubes (male and female connector), Washing Solution and Storage Solution bottles have only one.
  • Page 56 4 Connect the 4-end fluid sensor cable to the port labeled Bottle Sensor at the back of the instrument (Figure 1.2) and fasten securely. 5 Note the color coding and connect each fluid sensor cable to the respective bottle closure. 6 Connect a hydrophobic air filter to the appropriate connectors on the bottle closures to keep buffers sterile.
  • Page 57 1 Gently insert the Single tube rack into the corresponding slots located at the front of the instrument. 2 To remove the Single tube rack, hold it between two fingers and pull gently. Install the MACSQuant X Orbital Shaker Refer to the MACSQuant X Orbital Shaker user manual for details on its installation.
  • Page 58 Connect the instrument to the power supply 1 Connect the power cord to the power socket at the rear side of the instrument. The main power switch is located on the right side of the instrument. Ensure that the main switch is in position O (off) before connecting the power cord.
  • Page 59 Operating the instrument Switching on the instrument 1 Use the main switch on the right-hand side of the instrument to switch the instrument to standby mode. A red LED in the lower left corner of the screen indicates that the instrument is in standby mode. 2 Tap the screen to start the MACSQuantify Software.
  • Page 60 Needle priming time: defines the priming time when a new experiment is started to avoid that the needle runs dry (20 seconds are recommended) 3 Go to Shutdown default behavior. 4 Select Instrument off or Data analysis mode as the default behavior for all users. Long-term storage The instrument must be run every two weeks in order to prevent clogging.
  • Page 61 MACSQuant Instrument, the user must be familiar with the MACSQuantify Software. It is highly recommended that hardware calibration is only performed by an administrator who has been trained by Miltenyi Biotec. Refer to the MACSQuantify Software user manual or the help function of the MACSQuant Instrument for an introduction to the MACSQuant Instrument user interface and a detailed description of the instrument settings.
  • Page 62 3 Manually adjust the robotic needle arm in order to position the uptake needle over the center of the port. 4 Gently lower the robotic needle arm manually into the port until the uptake needle touches the bottom of the port. Do not bend the needle or insert it diagonally. 5 Click Save calibration.
  • Page 63 If the MACSQuant X 5 Rack is not selected, the MACSQuant Instrument will display a prompt screen. 4 Place the MACSQuant X 5 Rack onto the MACS Orbital Shaker and place an empty 5 mL tube in position D6.

  • Page 65 13 Click Test to confirm successful calibration. The MACSQuant Instrument tests all positions. Calibrating the Single tube rack detection Activation of the Single tube rack detection avoids a collision of the MACSQuant X Orbital shaker with a mounted Single tube rack.
  • Page 66 3 Follow the instruction on the screen. 4 The closed green circle indicates that the calibration is completed. After all calibrations are completed, the instrument can be switched into acquisition mode. 4.10 Volume calibration After exchange of the sample tubing, the system must be calibrated to this specific tubing in order to correctly set the tube volume and flow rate.
  • Page 67 11 Select Continue. 12 After the volume calibration has completed, the status of the calibration is indicated in the upper right corner. Passed indicates that the calibration was successful. Otherwise Failed indicates that the new tubing could not be identified correctly. In this case, repeat the calibration. If problem persists, contact Miltenyi Biotec Technical Support.
  • Page 68 7 Place an empty 5 mL tube in the Single tube rack. 8 Click the Start button. 9 Follow the on-screen instructions. 10 Select Continue. 11 After the tube length calibration has completed, the status of the calibration is indicated in the upper right corner.
  • Page 69 The Sample uptake unit tab indicates the status of the robotic needle arm as well as whether the MACS MiniSampler Plus or the MACSQuant X Orbital Shaker is connected or not. The relative position of the robotic needle arm is listed in the lower left box as well as whether the Single tube rack is connected or not.
  • Page 70 The operating temperature for the main laser should be between 35 °C and 45 °C. The main laser is the blue laser for MACSQuant X. The side lasers are kept at a temperature of 25 °C. When in acquisition mode, colored lines from the lasers to the detectors indicate that lasers are switched on.
  • Page 71 Live Support MACSQuant Live Support is a real-time diagnostic service provided by Miltenyi Biotec Technical Support. Highly trained MACSQuant Specialists can be reached in real-time to assist with any queries you may have. A webcam is provided for communication during MACSQuant Live Support, which is attached via USB connection to the MACSQuant Instrument.
  • Page 73 To avoid clogging due to deposition of dust, exchange filters once a year. Use only the hydrophobic air filters from Miltenyi Biotec (# 130-090-385). 1 To exchange the air filter of the fluidics system, remove the Single tube rack if installed.

  • Page 75 3 Lift the needle upwards to remove from the needle sealing. 4 Unscrew the knurled screw of the sample tubing. Do not lose the inlay. 5 Remove the sample tubing and replace it with a new tubing. 6 Insert the inlay into the needle holder.
  • Page 76 7 Tighten the knurled screw. 8 Insert the needle back into the slide in the z-axis. Do not bend the needle. 9 Snap the needle holder back into the needle arm. 10 As the needle has been removed from the needle arm, its position might have been changed. Check the position and recalibrate if necessary.
  • Page 77 3 Press the strap latch (orange arrow) to release the strap. 4 Lift the strap.
  • Page 78 5 Remove the tubing strap. 6 Disconnect the needle from needle arm. Refer to Exchanging the uptake needle on page 72. Take care not to bend the needle. 7 Unscrew the knurled screw of the sample tubing. Do not lose the inlay or the stopper sleeves. Remove the sample tubing and discard it.
  • Page 79 16 Press the right upper corner until the strap snaps back into position. 17 After exchange of the sample tubing, the system must be taught to this specific tubing in order to correctly set the tube volume and flow rate. This process is automated and ensures correct cell counts. Start the Express Mode VolumeCalibration to start the process.
  • Page 81 In case of leakage of the syringe pump, turn off the instrument and check the connection between the syringe pump and the valve. If it is loose, tighten it counterclockwise. If the leakage persists, contact Miltenyi Biotec Technical Support to replace either the pump syringe or the pump seal.
