4 Click Start calibration under the heading Chill 384 rack. The robotic needle arm will automatically move
forward.
5 Adjust the uptake needle to the center of the well P24 (i.e. equidistant from the well edges). Move the
needle arm forward or backward. Move the MACSQuant X Orbital Shaker for movement to the right and left
side.
6 Lower the robotic needle arm carefully until it touches the bottom of the well.
7 Click Save calibration. The closed green circle indicates that the calibration is completed.
8 Select test position in the rack window in the side panel.
9 Click Test to confirm successful calibration. The MACSQuant Instrument tests selected positions.

Calibrating the MACSPlex Filter Plate
1 Go to the Tools tab in the side panel of the MACSQuantify Software and select the Calibrate uptake unit
box. Red closed circles indicate that the uptake components are not calibrated.
2 Go to the Experiment tab in the side panel and select MACSPlex Filter Plate from the Rack drop-down
menu. If the MACSPlex Filter Plate is not selected, the MACSQuant Instrument will display a prompt screen.

3 Place an empty MACSPlex Filter Plate on the MACS Orbital Shaker.
4 Click Start calibration under the heading MACSPlex Filter Plate. The robotic needle arm will
automatically move forward.
5 Adjust the uptake needle to the center of the well H12. Move the needle arm forward or backward. Move
the MACSQuant X Orbital Shaker for movement to the right and left side.
6 Lower the robotic needle arm carefully until it almost touches the well bottom (i.e. only a fraction of a
millimeter from the bottom of the well).
7 Go to the field Z-offset and enter a value of -7.
8 Press enter.
Be careful not to damage the membrane of well H12 when calibrating the MACSPlex Filter Plate.
9 Click Save calibration. The closed green circle indicates that the calibration is completed.
10 Select test position in the rack window in the side panel.
11 Click Test to confirm successful calibration. The MACSQuant Instrument tests selected positions.

Calibrating the custom 96 and custom 384 racks
If 96 and 384 plates of different format (e.g. deep well or round bottom) are used, they might have a different
distance from the bottom of the MACSQuant X Orbital Shaker to the bottom of the well. In order to pick the
samples correctly, individual "custom" racks can be defined for each required rack format.
1 Go to the Tools tab in the side panel of the MACSQuantify Software and select the Calibrate uptake unit
box. Red closed circles indicate that the uptake components are not calibrated.
2 Go to the Experiment tab in the side panel and select Custom 96 Well Rack or Custom 384 Well Rack
from the Rack drop-down menu. If the Custom Rack is not selected, the MACSQuant Instrument will display
a prompt screen.
