5 Click Save calibration. The closed green circle indicates that the calibration is completed. Click Test to
confirm that the correct coordinates are saved.

Calibrating the MACSQuant X 5 Rack
For the correct usage of the MACSQuant XOrbital Shaker, calibrate the MACSQuant X 5 Rack.
1 Attach the MACSQuant XOrbital Shaker to the MACSQuant Instrument. Refer to the MACSQuant X Orbital
Shaker user manual for details.
2 Go to the Tools tab in the side panel of the MACSQuantify Software and select the Calibrate uptake unit
box. Red closed circles indicate that the uptake components are not calibrated.
3 Go to the Experiment tab in the side panel and select MACSQuant X 5 rack from the Rack drop-down
menu. If the MACSQuant X 5 Rack is not selected, the MACSQuant Instrument will display a prompt screen.
4 Place the MACSQuant X 5 Rack onto the MACS Orbital Shaker and place an empty 5 mL tube in position D6.
5 Click Start calibration under the heading Chill 5 rack. The robotic needle arm will automatically move
forward.
6 Adjust the uptake needle to the center of the tube in position D6 (i.e. equidistant from the tube edges).
Move the needle arm forward or backward. Move the MACSQuant XOrbital Shaker for movement to the
right and left side.
7 Lower the robotic needle arm carefully until it touches the bottom of the tube.
8 Click Save calibration. The closed green circle indicates that the calibration is completed.
9 Select test position in the rack window in the side panel.
10 Click Test to confirm successful calibration. The MACSQuant Instrument tests selected positions.

Calibrating the Chill 96 Rack
1 Go to the Tools tab in the side panel of the MACSQuantify Software and select the Calibrate uptake unit
box. Red closed circles indicate that the uptake components are not calibrated.
2 Go to the Experiment tab in the side panel select the Chill 96 rack from the Rack drop-down menu. If the
Chill 96 Rack is not selected, the MACSQuant Instrument will display a prompt screen.
3 Place an empty 96-well plate on the MACSQuant X Orbital Shaker.
4 Click Start calibration under the heading Chill 96 rack. The robotic needle arm will automatically move
forward.
5 Adjust the uptake needle to the center of the well H12 (i.e. equidistant from the well edges). Move the
needle arm forward or backward. Move the MACSQuant X Orbital Shaker for movement to the right and
left side.
6 Lower the robotic needle arm carefully until it touches the bottom of the well.
7 Click Save calibration. The closed green circle indicates that the calibration is completed.
8 Select test position in the rack window in the side panel.
9 Click Test to confirm successful calibration. The MACSQuant Instrument tests selected positions.

Calibrating the 384-well plate
1 Go to the Tools tab in the side panel of the MACSQuantify Software and select the Calibrate uptake unit
box. Red closed circles indicate that the uptake components are not calibrated.
2 Go to the Experiment tab in the side panel of the software and select the 384-well format from the Rack
drop-down menu. If the 384-well is not selected, the MACSQuant Instrument will display a prompt screen.
3 Load an empty 384-well plate onto the MACSQuant X Orbital Shaker.
