There are no obvious signs that indicate contamination and/or
instability of the probe.
Storage conditions other than those specified above must be verified
by the user.
Troubleshooting
Pretreatment (overdigestion): The Instructions For Use of the BOND
Enzyme Pretreatment kit (AR9551) recommends a dilution of 1 drop of
Enzyme Concentrate 1 into 7 ml of Diluent to be registered as *Enzyme
1 in the BOND software.
This *Enzyme 1 may be too strong for certain specimens when used
in combination with the Heat Induced Epitope Retrieval (HIER) and
results in overdigested tissue. A secondary dilution of the *Enzyme
1 (up to 1:300 dilution) is recommended and a separate registration
of the reagent on the BOND system as *Enzyme 5 is recommended
to prevent confusion. Alternatively, a direct dilution of up to 1:3000
of Enzyme concentrate 1 can be used instead of the serial dilution of
*Enzyme 1.
Probe dilution and Signal intensity: A 1x final probe concentration
is recommended for use but optimal probe balance is dependent on
factors such as tissue age, tissue fixation, pretreatment protocol, and
personal preference. Optimal concentrations and probe ratios may
vary and are to be determined empirically by the end-user.
