TEST PROCEDURE
Important procedural notes!
• Do not interchange reagents from different
kit lots.
• Reagents and samples should be used
equilibrated to room temperature (20-25°C),
approx. 30 minutes in opened box.
• Always mix conjugate before use, by inverting
the bottle twice.
• The procedure steps should be performed
without any delay.
• The kit can be stored at room temperature
for one week.
Sample material
Citrated platelet-free plasma.
Sample preparation
Preparation of plasma
Venous blood is drawn in evacuated blood
collecting tubes for coagulation analyses (0.11
or 0.13 M Na-citrate). Alternatively, 1 volume
of 0.11 or 0.13 M Na-citrate is added to 9
volumes of whole blood in a tube.
Centrifuge samples for 15 minutes at 2000 g.
Important
Remove plasma from the upper part of the
tube using a pipette. The plasma should not
be poured off. Avoid contamination with cells.
The sample should be centrifuged within 6
hours, preferably immediately after sampling.
The plasma should be tested within 24 hours.
Frozen samples
The plasma samples can be kept frozen at
-20°C for 4 weeks. Thawing of plasma samples
should be performed in a waterbath at 37°C
for 15 minutes. An additional centrifugation is
recommended. See "Troubleshooting".
Lyophilized samples
Lyophilized samples will give flow problems.
These samples should be centrifugated or sterile
filtrated before use. See "Troubleshooting".
Internal quality control
The positive control should be used to confirm
the efficacy of the reagents and the correct
performance of the test. The measured value
should be within the acceptable limits stated
on the vial label.
1 Prewashing
Apply 50 µL of washing solution to the test
device. Avoid touching the membrane with
the pipette. Allow the washing solution to
soak into the membrane.
2 Sample
Apply 50 µL undiluted platelet-free citrated
plasma or control to the test device.
The sample should be absorbed into the
membrane in less than 50 seconds.
3 Conjugate
Apply 50 µL of conjugate to the test device.
The conjugate should be absorbed into the
membrane in less than 50 seconds.
4 Washing
Apply 50 µL of washing solution to the
test device.
