The use of this semi-quantitative method is also described in
Appendix C of the EN ISO 15883-1 standard.
Reagent A reacts with proteins to form a Cu
complex.
With reagent B the Cu
+
reduced to Cu
, which reacts with reagent C to give a purple
complex.
The concentration of this complex is inversely proportional to
the amount of protein present.
With this method a protein content of between 10 µg and
60 µg protein/ml is detectable using the reflectometer.
Three levels can be assessed optically using the colour
comparison chart, <30 µg, 30-60 µg and ü60 µg.
This means that the limit of determination when using 2 ml
SDS solution for the assay is approx. 20 µg protein or when
using 3 ml SDS solution approx. 30 µg protein for the total
test volume.
