Introduction
2.1 Intended use
The MACSxpress Separator is a laboratory equipment for manual separation of
magnetically labeled cells. The device contains a permanent magnet for cell
separation and uses LEDs with a rechargeable battery for illuminating of the
target cell fraction. The MACSxpress Separator is used for untouched isolation
of leukocytes from blood products without density gradient centrifugation.
Erythrocytes are sedimented, while non-target cells are removed. Up to 30 mL
of prepared sample can be processed at a time. The instrument is for research
use only and not for therapeutic or diagnostic use. It is to be used by
professional laboratory personnel only.
2.2 Separation principle
The MACSxpress Separator is developed for the untouched isolation of
leukocytes from up to 30 mL anticoagulated whole blood without density
gradient centrifugation. Erythrocytes are aggregated and sedimented, while
non-target cells are removed by immunomagnetic depletion with
MACSxpress Beads. The tube containing the labeled sample is placed in the
magnetic field of the MACSxpress Separator. The magnetically labeled cells
will adhere to the wall of the tube while the aggregated erythrocytes
sediment to the bottom. While the tube is still inside the MACSxpress
Separator, the supernatant containing the non-labeled target cells can be
carefully pipetted into a new tube (
